My new laswer kit says to assemble the walls up to a point, then paint or stain a wall as it will not be practical to do so after more assembly. I have been using white glue so far and it works well but will it still work to further assemble the remaing , painted, WALLS,?

It may. However, using one of the alphatic[sp] resin glues like Titebond will work better on painted parts. Better yet would be gap-filling CA, although it might help to hit the painted surfaces to be bonded with a little sandpaper first.
Goo and contact cements will also work, especially when bonding dissimilar materials.

It will probably work as well on painted or stained walls as on bare wood. However, I would recommend some sort of carpenter's glue that is not water soluble even after it sets.
No matter what type of glue you use, if you plan to stain the wood (as opposed to painting it) you should stain it before assembly. Wherever there is any excess glue (even where you wiped it off while still wet) the stain will not be fully absorbed into the wood as it will in the other areas.
