Forgive me if this is buried somewhere back in the threads - a couple of searches didn't get me anywhere.

I'm trying to duplicate a specific line (Adirondack between Lake Placid and Saranac Lake). I have enough information on the topography to let me come reasonably close to the profile (at least close enough for my tastes at the moment), but I'm having trouble getting them smoothed in. The "slope" option is giving me fits. Since the grades are a significant part of the line (there is a section of 2%), getting this right is important.

I started on the LP end - high point of the profile. Should I have started at the lowest point and worked uphill? Is there a way that I've missed to set the elevation of the lowest and highest points of the slope?

I am not sure I can help you much with your problem although I can create a very good grade when I build my layouts with Trainz. Starting from the top or bottom shouldn't make any difference but one of the keys to making it work is not using too many spline points. If possible keep the track one single line then use the 'adjust height' button in surveyor. You can certainly go to the trainz forum and under 'Route Builders' and I am sure you will get all the help you need.

My real reason for this response, is the area you are planning to duplicate. My family has a cottage in that area. I have spent many years (and still do when I get the chance) there. It is located in Mountain View about 20 mile or so from Malone. I have been to Lake Placid many times while there and have loved the place. Our cottage was bought by my Grandfather in 1947 while he was working for CP - out of the Farnham Yard in Quebec. His route took him through Upper New York State. At one time that was how we use to get to Mountain View and I vividly remember the hauting sounds of the train whistle as it echoed off the Mountains in the early evening.
