I try to buy everything at my LHS, but I still do a lot of browsing.  (Then, I order it through the LHS.  He gives me a discount, and there's no shipping.  Great for small things.)

My two favorite browsing sites, because of their large catalogs, on-line pictures and search capabilities, are:

www.walthers.com and www.discounttrainsonline.com.

I agree with online businesses that Glenn has listed above.

Except for the occasional kit on eBay, I only buy my locomotives online.  Pretty much everything else I get from my LHS.  I can view and inspect the product, delivery is prompt, and there's no shipping cost.  (Although, it's probably figured into the price somehow.)  I know that for some folks, an LHS is not always convenient or accessible, and they have to reply on either catalog or online businesses to purchase their supplies from.

What are the best websites or your favorites website for making purchases.  Ease in navigation, opportunity to view products, prompt delivery, and competitive prices are all important.
