I have never tried this method. I have read about it over the years since Bill McClanahan wrote about it in his book called Scenary for Model Railroads that came out in the late sixties . It was used to treat raw lichen that came in blocks from Walthers or wild lichen picked in the field. I could never find glycerin and didn't know where to get it in the town I lived in. It was like hydrocal, something of a legend,  heard of but never experienced directly. You can get lichen in large batches from scenery express already treated. I would just go that route.

Does Glycerine soften Lichen?

It did for me.  I have a bag full of lichen that I picked 40 years ago, first drying it, then soaking it in glycerine.  It's still soft to this day.
